SUBSTRATE OPTIMIZATION FOR EXOPOLYSACCHARIDE PRODUCTION BY RHODOTORULA MINUTA BIOTECH 2178 USING SIMPLEX-LATTICE DESIGN

Rhodotorula minuta BIOTECH 2178 are known exopolysaccharide (EPS)-producing yeast
having complex characteristics potential for biotechnological application. Formulation
optimization following Simplex-Lattice Design (SLD) was carried out to investigate the
combined effect of the three relevant variables, namely: levels of yeast extract, glucose,
and xylose to enhance production of exopolysaccharide (EPS) using Rhodotorula minuta
BIOTECH 2178. The optimal substrate formulation for EPS production was found at
18.75g/L yeast extract and 6.25g/L xylose. The superimposed plot revealed that glucose did
not have a significant effect on the optimum EPS production. Statistical analysis revealed the
regression model was in good agreement with the experimental results with a determination
coefficient (R2
adj) of 0.982706. Under these conditions, the maximum predicted EPS yield
was 2.11g/L. The total sugar present in EPS was 1.07g/L, which was 2.5-fold greater,
compared to the EPS that was produced during variable screening. Verification studies
showed no significant difference between the actual and predicted values of the responses.
These findings revealed the adequacy and applicability of Response Surface Methodology
using SLD for substrate optimization for EPS production using R. minuta BIOTECH 2178.
